The pioneering Sisters—Mother Ephrem Marie Fieu, Sr. Suzanne du Sacre Coeur Cran, Sr. Agnes de Sainte Anne Couplan, Sr. Jeanne de St. Louis Bourrelly, Sr. Marie Angeline Acau, and Postulant Sr. Ste. Foy de Sacre Coeur Sacramento—responded to an invitation from Bishop Dennis Dougherty to establish a school in Cagayan Valley. The institution’s name evolved over time: from Colegio del Sagrado Corazon de Jesus (1909) to Sacred Heart of Jesus Institution (SHOJI) (1925). In 1934, the community moved to the Colegio de San Jacinto grounds (where the University stands today) after acquiring it from the Dominican Order.

During World War II, the school served as a military garrison and hospital for the Japanese forces. Despite challenges, SPUP emerged resilient, assuming the name St. Paul College of Tuguegarao (SPCT) in 1948 and becoming the first Teacher-Training Institution in Cagayan Valley. In 1961, it became the first accredited institution in the region by the Philippine Accrediting Association of Schools, Colleges, and Universities (PAASCU). The University faced adversity again when the entire complex was reduced to ashes in 1965. Undeterred, SPUP achieved a significant milestone in 1982 by becoming the first University and Catholic University in the Cagayan Valley region.
